    A Novel Molecular and Cellular Study on Curcumin

    Since the discovery of G-quartet (G4)by M. Gellertin 1962, much attention has been given on G4and C4(also called i-motif) as important drug design targets for the treatment of various human disorders. G4 forming sequences are prevalent in human genome, which includes many important regions of the eukaryotic genome, such as telomere ends, regulatory regions of many oncogenes c-kit, proto-oncogene c-myc, Kirsten rat sarcoma viral oncogene homolog (KRas). Curcumin(diferuloylmethane), an antiinflammatory and antioxidant compound, is found in the rhizomes of the plant Curcuma longa. The phytopolyphenolic chemical curcumin has been in the prominence due to its diverse pharmacological activities. Here, we studied the binding of curcumin with G-quartet and duplex DNA as well as protein bound DNA. Curcumin showed inclination toward binding with G4 than C4 and duplex sequences. Furthermore, cellular studies have been initiated on HeyA8 ovarian cancer cells. Curcumin treatment inhibited the proliferation of HeyA8 cells in a dose responsive manner demonstrated by MTT assay. studies on the binding of curcumin with HeyA8 DNA are underway

    Evaluation of the Feeding Value of Proso Millet in Growing-Finishing Diets for Pigs and Effects of Feed Ingredients and Medium-Chain Fatty Acids on Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) Survivability

    The objective of the project were to: 1) evaluate the feeding value of proso millet as a potential substitute for corn in corn-soybean meal-based diets for pigs; and 2) investigate the relationship between feed ingredients and virus survivability, followed by examining feed additive candidates that could mitigate the risk of transmission of viral pathogens. In Exp. 1, 36 barrows were fed 1 of 4 dietary treatments, including: Diet 1, corn-soybean meal-based (control), and Diets 2, 3, and 4 had proso millet replacing 33%, 67%, and 100% of corn in the control diet, respectively. During Phase 3, pigs consuming the proso millet diets (Diets 2, 3, and 4) had greater ADFI than pigs consuming the basal diet, but ADG:ADFI was not different among groups. The growth parameters (i.e., ADG, ADFI, and ADG:ADFI) were not affected by treatments during the whole experimental period (from Phase 1 to Phase 4), suggesting that the level of corn replacement up to 100% by proso millet did not affect pig growth. In Exp. 2, ground corn, SBM, DDGS, and a complete feed, were inoculated with PRRSV, followed by a cell-based antiviral assay and qRT-PCR to evaluate the infectivity of each virus-inoculated ingredient. A significant reduction of infectious PRRSV was observed in ground corn, DDGS and complete feed, while SBM retained the highest amount of the infectious virus among ingredients and controls. The results suggested that the survivability of PRRSV in feed appears to be influenced by ingredient type. The follow-up experiment was conducted to assess the antiviral activity of selected MCFAs, as determined by antiviral assay and qRT-PCR. The antiviral assay showed that all tested MCFA and MCFA blends inhibit PRRSV in a dose-dependent manner, indicating that MCFAs and the specific mixture of fatty acids may potentially serve as mitigants for reducing the risk of PRRSV transmission via feed.     Implementation and Verification of Three-Dimensional MOC Transient Solver in Whole-core Transport code STREAM

    Department of Nuclear EngineeringIn the state of the art of computer technology, the transient analysis in the nuclear reactor has become on demand of the nuclear engineering field. This thesis presents the development and the preliminary validation of the transient transport capability in STREAM code. The Theta method with the well-known Crank-Nicholsen scheme providing a second-order accurate is then applied to tackle the time integration in the right-hand side of the time-dependent neutron transport. Eventually, the Methods Of Characteristic (MOC) solver in the steady state with excellent performance and accuracy in STREAM is modified with the delayed neutron term to solve the transient cases. Additionally, a multi-group Coarse Mesh Finite Difference (CMFD) accelerator is introduced to alleviate the computational burden of the simulation. The transient problems, namely TD0, TD1, TD2, TD3, TD4 and TD5 in the C5G7-TD benchmark suite are used for verification. The STREAM time-dependent MOC calculation results show good agreement with results of a deterministic transport analysis code, nTRACER with the maximum disparity of the total power level change is 1.22 % for the TD3-4 case. In the 3D cases, with a proposed decusping method, the maximum amplitude of the relative error is about 3.5% between the two deterministic codes. The disparities induced by decusping methods and whole-core transport method are obviously observed and thus are the primary source causing the discrepancies. With this high fidelity to replicate the solution in the time-dependent transport equation, the transient analysis capability of STREAM code has been proved. This work plays as a foundation for the later 3D whole core transient solver accompanied by TH1D feedbacks for practical transient problems.ope
